Combining Mabtech's trusted monoclonal antibody pairs with magnetic beads, EYRAplex assays are designed to deliver reliable results every time. From focused 6-plex panels to broad 30-plex formats, EYRAplex lets you analyze more with less saving you time, resources, and sample volume.

The same, highly specific mAb pairs used in our ELISpot, FluoroSpot, and ELISA assays are now available in a new multiplex format with EYRAplex. And with our recombinant mAbs and novel assay buffer, heterophilic antibody interference is a thing of the past. Keep reading to learn more!

How it works:

Magnetic bead-based multiplex assays have been trusted for years to help researchers detect multiple analytes in solution saving time and resources. We've simply made them better. The approach immobilizes capture antibodies onto magnetic beads with a dye ratio specific to each analyte. These capture beads are pooled together as a panel and added to a 96-well plate. Samples and standards are added where analytes are captured during incubation. The excess sample is washed away and biotinylated recombinant detection antibodies are added. The final addition of a streptavidin-PE conjugate allows for measuring the amount of captured analyte. Mean fluorescent intensity (MFI) values from data acquisition are converted into analyte concentrations based on standard curves.

EYRAplex assays have been tested and validated for use with plasma (heparin or EDTA), serum, and cell supernatants.

Bead IDs: the key to multiplexing

At the core of magnetic bead-based multiplexing lies the Bead ID. In EYRAplex assays, each magnetic bead is dyed with a unique ratio of fluorescent dyes, creating a distinct Bead ID. A specific capture antibody is immobilized on each bead type, linking the Bead ID directly to a single analyte. This elegant design enables true multiplexing allowing all analytes to be measured simultaneously in a single well using a shared PE detection signal. Mabtech EYRA™ with the RAWsphere algorithm identifies the Bead ID and quantifies the PE signal intensity, easily matching each fluorescent event to its correct analyte.

Goodbye heterophilic antibody interference

When we started on our assay development journey, we looked to current multiplex assay offerings and the issues they were facing. We quickly discovered that many assays had major issues with heterophilic antibody interference. Heterophilic antibodies, including human anti-mouse antibodies (HAMA), commonly interfere with assays, leading to false positives by cross-linking capture and detection antibodies or false negatives by blocking one of them. This issue increases in autoimmune diseases and infections, i.e., clinical samples. EYRAplex assays address this by using recombinant detection mAbs and a specialized assay buffer to block interference giving you the most accurate data possible.

Results from benchmarking tests against Competitor 1 by spiking blank samples with rheumatoid factor (RF). The RF spike significantly increased the false positive signal in Competitor 1's magnetic bead-based multiplex assay compared to EYRAplex.
