Intended use

This ELISA Pro kit is intended for the quantification of human latent Transforming Growth Factor-β1 (TGF-β1) in serum, plasma, and cell culture supernatants. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

The ELISA diluent included in this kit blocks heterophilic antibodies from cross-linking the assay antibodies, thereby preventing false-positive read-outs. Heterophilic antibodies are commonly found in human serum/plasma and can also be present in other species. The ELISA diluent has been validated using serum/plasma samples from healthy human blood donors. For quantification of latent TGF-β1 in blood, the use of plasma is recommended since serum contains high levels of latent TGF-β1 released from platelets during sample preparation. Plasma can be obtained using EDTA, citrate or heparin as anti-coagulants. To minimize the platelet content in the sample, an additional centrifugation of plasma at 10,000 x g for 10 minutes is recommended.

Quantification of latent TGF-β1 - no need for sample pre-treatment
TGF-β1 is secreted in a latent, biologically inactive form, non-covalently linked with a Latency Associated Protein (LAP). The monoclonal antibodies MT593 and MT517-biotin bind to the LAP entity of human latent TGF-β1. This enables direct quantification of latent TGF-β1 in ELISA with no need of sample pre-treatment.

This kit is based on a matched pair of mAbs specific for the Latency Associated Protein (LAP) of human latent TGF-β1.

Cross-reactivity

The ELISA does not detect human latent TGF-β2 or -β3 or bovine latent TGF-β1. The mAbs cross-react with latent TGF-β1 from non-human primates (NHP). For more information please check the Cross-reactivity guide section below.

Transforming growth factor β1 (TGF-β1) is secreted by many cells and controls functions such as cell growth, cell differentiation, proliferation, and apoptosis. The cytokine TGF-β1 is an important regulator of immune functions. Specifically for T cells, TGF-β1 promotes the differentiation of regulatory T cells, preventing IL-1- and IL-2-dependent proliferation of activated T cells, the activation of helper T cells, and cytotoxic T cells. TGF-β1 is secreted in a latent, biologically inactive form, non-covalently linked with a latency associated protein (LAP).
