Intended use

This monoclonal antibody enables specific detection of human apoE in immunoassays such as ELISA, ELISpot and Western blot. The antibody is also suitable for immunoprecipitation.

Recommendation

E887 is recommended as detection mAb in ELISA and ELISpot in combination with capture mAb E276 (product code 3712-3) for human apoE. For monkey apoE, E887 should be used in combination with capture mAb E981 (product code 3712M-3).

Specificity

Native and recombinant human apolipoprotein E (isoforms apoE2, apoE3, and apoE4). The presence of low amounts of detergent is necessary for binding. Tween 20, Triton X100 or NP40 can be used at a concentration of 0.01-0.5%. Do not vortex with detergent added.

Cross-reactivity

Cross-reacts with native apoE from rhesus macaque and cynomolgus macaque. For information on reactivity with other non-human primates (NHP), please check the NHP cross-reactivity guide.

Purification

Purified from in vitro cultures by protein G affinity chromatography.

Biotinylation

Biotinylated through reaction with a N-hydroxysuccinimide ester of biotin.

Detection of human apoE with mAb E887 in Western blot. Human plasma (0.1 µl and 0.01 µl) in SDS sample buffer were denatured at 70˚C for 10 minutes and run on SDS-page / Western blot. ApoE was detected using 1 µg/ml E887 (biotinylated) followed by SA-ALP and BCIP/NBT-plus substrate.

The systems reactive with NHPs are either based on cross-reactive human kits or specifically developed monkey kits (NHP).

Cross-reactivity verification tests have been performed in ELISpot and/or ELISA by Mabtech and/or by others. Evaluations of assays with less solid evidence of cross-reactivity are shown as symbols within parentheses.

Analyte description

Apolipoprotein E (apoE) is a 34 kD glycoprotein involved in lipid transport, lipoprotein metabolism, and cellular signaling. It is present in plasma lipoprotein particles such as chylomicrons, VLDL, and HDL. ApoE is also present in large amounts at sites of neurological damage and appears to be involved in the recycling of apoptotic remnants and amyloidal aggregates. ApoE is mainly produced in the liver and by macrophages, but also by other cell types such as smooth muscle cells or neuronal cells. In humans, apoE exists in three isoforms, apoE2, apoE3, and apoE4. Of these, apoE4 is associated with Alzheimer’s disease. A normal concentration of apoE in human plasma is approximately 30-40 µg/ml. The concentration of ApoE in inbred mouse plasma can vary between 15-150 µg/ml.
