Intended use

This ELISA Flex kit is intended for the quantification of human IL-21 in solution. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

Analysis of serum/plasma with this kit requires the use of ELISA diluent. The ELISA diluent blocks heterophilic antibodies from cross-linking the assay antibodies, thereby preventing false-positive read-outs. Heterophilic antibodies are commonly found in human serum/plasma and can also be present in serum/plasma from other species. The ELISA diluent has been validated using serum/plasma samples from healthy human blood donors. It should be used for dilution of standard, samples, and detection antibody.

Detection of IL-21 in cell culture supernatants
To retain detectable levels of IL-21 in cell culture supernatants, biotinylated mAb MT21.3m can be included during cell culture. The antibody will bind to secreted IL-21 thus preventing IL-21 from interacting with its receptor. In this way, IL-21 is kept in solution and can be quantified in ELISA. For this purpose, mAb MT21.3m is available as a separate reagent, i.e., not included in the kit, free of preservatives.

IL-21

Analyte description

Interleukin 21 (IL-21) is primarily secreted by CD4 T cells (particularly follicular T cells and Th17) and is an important regulator of immune responses. For Th17 cells, IL-21 is an autocrine regulator sustaining Th17 development. IL-21 also regulates the development of other T cells as well as B cells, NK cells, and dendritic cells. Areas of particular interest for research on IL-21 include autoimmune and inflammatory diseases, cancer research, and basic studies on, e.g., B cells and T cells, especially on Th17 cells.
