Intended use

This ELISpot Flex kit is intended for the enumeration of cells secreting human IgE using the ELISpot assay. For research use only. Not for use in diagnostic procedures.

Serum/Plasma samples

IgE secretion can be induced by pre-activation of cells with the included anti-CD40 mAb and IL-4. Please note that stimulation with anti-CD40/IL-4 induces switching to IgE and IgG1 and results with stimulated cells may therefore not be representative of the situation in vivo.

Shipping and Storage

Shipping

Shipped at ambient temperature.

Storage

Store capture mAb, detection mAb, and enzyme conjugate at 4-8 °C upon receipt. Anti-CD40 mAb and IL-4 should be stored at -20 °C.

Shelf lifeAt least 18 months from date of receipt.
PBMC were pre-incubated for 5 days in the absence or presence of anti-CD40 monoclonal antibody and recombinant IL-4. The cells were then washed, added into ELISpot wells and analyzed for the secretion of total IgE (250,000 cells/well, overnight incubation).

Selecting the appropriate mAbs is key to detecting both total and antigen-specific human IgA. This blog post explores Mabtech’s mAbs, tailored for ELISpot to detect human IgA antibody-secreting cells.

IgE

Analyte description

The main physiological function of immunoglobulin E (IgE) is to protect against parasitic infestation. However, IgE is also associated with allergy, and allergen-specific IgE antibodies may trigger allergic reactions in the presence of the allergen. IgE is normally found in low amounts in serum/plasma but is significantly increased in allergic individuals, and the detection of allergen-specific IgE serves as the basis for many in vitro assays for allergy diagnosis.
