Intended use

This kit is intended for FociSpot immunostaining of the influenza B nucleoprotein, which enables enumeration of influenza B virus foci in focus forming assays (FFA). For research use only. Not for use in diagnostic procedures.

FociSpot
In FociSpot, virus foci are detected by using mAbs specific to viral proteins. After fixation and permeabilization of infected cells in a 96-well plate, the virus-specific biotinylated detection mAb is added. The use of Streptavidin-ALP enzyme conjugate, followed by a precipitating substrate enables detection of foci. Foci can be counted manually, but counting with Mabtech IRIS 2 is rapid and offers easy data handling.

FociSpot immunostaining of influenza B nucleoprotein in infected MDCK cells. MDCK cells were infected with influenza B virus (titrated, virus dilutions16,000x, 32,000x, and 64,000x are shown). After fixation and permeabilization, foci were detected using anti-influenza B (NP) mAb 4D5, followed by Streptavidin-ALP and BCIP/NBT-plus substrate. Foci were counted using the Apex software in Mabtech IRIS 2.

The Nucleoprotein (NP) is a structural protein and encapsidates the negative strand RNA of influenza B into ribonucleoproteins (RNP). NP plays a role in replication, transcription, RNA packaging, and nuclear trafficking. The NP does not only interact with the RNA, but also the viral polymerase, the M protein, and several host factors.
